If Windows 7 or Vista is formatted with MRB will it be able to read a USB external drive formatted GPT?

You don't format Windows. You format the drive Windows will be installed on. And you need to format any drives you expect Windows to access, but the drives do NOT need to be formatted with the same file system (NTFS, FAT32, etc).

Note the primary advantage to GPT is you can have more than 4 partitions on the disk.

Should be able to, but I haven't tested it.
