How can I lock down an entire drive so that no one can get access except me, when I need to ... Thank you

Several ways, but all put you data in more risk of being lost.

Software
1-encrypt the drive
2-hide the drive with permissions (and password)

Hardware
1-dongle without which the HD cant be accessed.
2-Physical means

But be careful forget a password, have a single glitch and you may lose it all
