Windows 7 shortcuts stop working

Ocassionally, some shotcuts on my desktop (not all of them) that refer to folders stop working unless I reboot. Then they work again. Any ideas on how to fix this?

Are the folders on a outside storeage drive (like a usb hdd) perhaps?

No, they are on the C drive.

Rebooting the system will dump the page files and that could help if the c drive is full... is this machine a vmware?
It may be worth doing a malware scan as well... we need more details to figgure it out.

No, it's not vmware. What other info would be useful? The OS is Win 7 64 bit.
