What is the easiest way to update Honeywell VM3 running on Windows 10 64 bit on 10240 build , i have around 200 device which need to be upgraded asap. I have tried build in windows update method which are failed when reached certain KB.

From what I read these run Windows 10 IoT Enterprise, if it's an enterprise edition you'd have to download the ISO from your MSDN licensing portal, if it's pro then you can download it from Link Removed. As for upgrading if you don't have some kind of patch management like SCCM then you're going to be manually upgrading them with a bootable usb drive.
